Yesterday, I tried to optimize my primer and PCR condition using V4 domain which is located in small subunit of mt-DNA but unfortunately it was not successful!? and I did not see anything in gel?!

So today I started to change the conditions including mgcl2, buffer, dNTPs, and...

I just hope...

After 2 weeks working hard, finally, I could extract DNA successfully. I found the problem but I do not discover the reason exactly. Around two weeks ago, the hypothesis just remained could be "my samples have problem", so, I went to my Mycology Lab and asked when I was working in Genetic Lab something wrong did not happen here??! The staff there told me, unfortunately, yes, happened. I realised that all my samples have demolished  because of changing the tempreture of incubator?!! Therefore, I started again and after 3 days I extracted DNA successfully.

Now a days, I am doing literature review on PCR reactions and DNA barcoding based on Mitochondrial DNA. I think from tomorrow I can start to work.

Today, I tried new CTAB method (modified CTAB method) but I did not run on the gel because there is not any time. I am going to run my samples tomorrow.

For many years, I have desired to work in Genetic, Microbiology, and Biotechnology Laboratories. Now, I think I have reached to my goal. From a few days ago, after doing my works in Mycology Lab succesfully, I started to work in my Genetic Lab. It was quite marvelous. "Oh, my god: DNA, PCR, Centrifuge, RAPD, Microsatellite, Mitochondrial and Ribosomal DNA" I said.

Hardworking again started. First, I must extract the DNA so I tried to get DNA from my tissue and spore cultures that I inoculated before. I used 4 different methods: CTAB, GF-1 Kit, Minikit for plants, and i-Genomic CTB methods. Then, I have run on the agarose 1% gel but unfortunately I have not still got the band! I do not know what is the problem?!

From tomorrow, I am going to work harder than the past until get the strong band. I will write the new post when I get the band. See you soon.

Importance of English

English is a language that everybody prefer to learn it because of a lot of advantages. First, it is universal and most of the countries around the world would like to use it as the second language. Second, It is easy to learn comparing to other languages like French, Persian, and Arabic. Third, for those who want to study abroad the best language will be English because most of the scientific books, journals, and papers are written in English. Because of these reasons, in my opinion English is going to be the best in the world.
